| 1. Microplate Indexing System |
This indexing system aligns the 8 pin replicator (87V-478) to appropriate
wells in a 96 well microplate. This provides a very simple method of placing
the replicator pins into the center of the wells by using a 4 set series of
guide
pins on the top of the indexing system and the pair of alignment holes in the
bottom of the Replicator. This registration system also is adjustable so y ou
can remove your specimen from the very bottom of your microplate or PCR
plat well, i.e., you can harvest from very small volumes even the dregs of the
well.
This indexer will accommodate all standard heght 96 well microplates that
conform to the
Society of Biomolecular Screening Standards. Many of the 96 well pp PCR
plates are smaller (length and width) than SBS Standard and need to be
shimmed for the correct registration. We provide shims for your convenience.
 | Eliminate the possibility of getting specimens from the wrong wells. |
 | Keeps the pins in the center of the well so the size of the spots remain
uniform.
This is very important especially in 384 well microplates. |
 | Keeps track of which set of wells you are spotting from and where the
next group of wells will be. |
 | Accelerates the rate of the arraying process. |
 | Prevents the pins from being damaged on the bottom and sides of the
wells.    |

2.A) FU-MEEI REGISTRATION SYSTEM
For "Xeroxing" blots onto membranes with MULTI-BLOT™ Replicators .
The Fu-Meei Registration System provides an efficient way of making identical
DNA blots on multiple small membranes
(xeroxing). It has its own built in three pin spindle system (87V- 440) for
registering the Replicator to the membrane that also
registers the blots to locating (reference/spindle) holes punched in the
membrane.
The locating hole features allows image analysis equipment convenient reference
features to locate and identify positive blots. Furthermore
our custom paper punch (87V- 441) is a convenient way to punch holes in many
nylon membranes (87V- 502) simultaneously
and place a hundred or more of them on the three pin spindle for blotting. The
spacing of the hole pattern also is used to orient
the user to location of the blots, right from left and top from bottom.
The Fu-Meei Registration System may also be used to make high density arrays by
using MULTI-PRINTS that have a 2 by 2 array
alignment pattern (87V-382F1) for 384 Pin MULTI-BLOT™ Replicators, a 3 by 3
array alignment pattern (87V- 382F2), and 4 by
4 array alignment pattern (87V- 382F3) for 96 Pin MULTI-BLOT™ Replicators. When
making high density arrays we recommend
that you use the 87V- 409 MULTI BLOT™ Replicator or 87V- 386 MULTI-BLOT™
Replicator.

2.B) VICKI ULTRA HIGH DENSITY ARRAY &
REGISTRATION SYSTEM
The Vicki Ultra High Density Array & Registration System provides a rapid fail
safe method of making 4 ultra high density
arrays from nine separate 384 well microplates (9 X 384 = 3456 individual
specimen blots) on a membrane. Each spot contains
~20 ul of specimen. The membrane can then be cut into 4 identical pieces and
processed independently or saved for later assays.
If you need to screen large DNA, cDNA, EST or RNA libraries this is the system
for you.
The operation procedure is the essence of simplicity:
 | A 87V- 381V Library Copier is placed over a 384 well microplate to
register the microplate to the Vicki Spring Loaded
MULTI-BLOT™ Replicator housing (87V- 386V) containing a 87V- 386VR1 Vicki
Replicator. The external edges of the Replicator
housing, index to an indentation in the LIBRARY COPIER™ thus assuring that
the pins are positioned over the center
of the wells. The spring handle is depressed lowering the pins into the
contents of the wells and the liquid is picked up on
the pin tips as ~20 ul hanging drops when the handle is raised. |
 | The external edges of the MULTI-BLOT™ Replicator housing indexes into a
window frame opening and rests on the membrane
while the Replicator pins are in the retracted position. The spring handle
is depressed causing the Replicator pins to simultaneously
blot the membrane. |
 | This process is repeated in all four window panels using the same source
plate and then the alignment pins are placed in the
next pair of alignment holes and the whole process repeated using a new 384
well plate. Multiple window frames may be used if
more than 4 identical blots are needed. |
 
The blot to the right was made from nine, 384 well
microplates (3,456 spots). Note that each set of 9
spots is distinct because of the spacing pattern between
pins. Each spot contains ~20 hl of specimen.
When using the Vicki Registration system you
have the freedom to choose the density of your
blots by simply selecting which of the 9 possible
blotting positions you use. |

Questions
| Q: How should I practice making arrays with
Replicators and the registration devices? |
A: Just add a dye (food color works fine) to the buffer, media or
solutions that you will be arraying. Do not put the dye into just
water as the surface tension of plain water is very high and will give
you variable results. A little DNA, protein or carbohydrate
makes a big difference. If your buffer doesn't or can't contain these
substances you can treat the pins with our 87V-110
Surfactant , or dip the pins in ETOH and flame them. |
| Q: When making arrays with the MULTI-PRINT™
should I use blotting paper under the membrane? |
A: Yes. We sell a wicking paper (87V- 521V 22cm x 32cm) that works
quite well to support the membrane for 4 MULTIPRINTS,
however Whatman, 3M or other similar paper will also work. |

The LIBRARY COPIER™ is a simple registration accessory that precisely aligns
the pins in our 48, 96, 384 or 1536 pin
MULTI-BLOT™ Replicators to the center of the wells in source and recipient
48, 96, 384 or 1536 well plates.
The single hole alignment pair is used on both the source and recipient
plates to copy the library. The ease and precision of the alignment ensures
a safe plate to plate transfer of cultures or clones, without "touching off"
the hanging drop. |

The COLONY COPIER™ is placed over a Nunc Omni Tray and aligns itself to the
external surfaces of the Omni Tray and precisely
registers the MULTI-BLOT™ Replicator's stainless steel pins to the interior of
the Omni Tray. High density offset arrays of blots or
colonies are produced on a single Omni Tray by inoculation at each alignment
hole pair using our unique system of guide pins and
alignment hole pairs.

libraries can be inoculated onto agar Omni Trays, grown into colonies and stored
in the refrigerator for 7 to 8 weeks. In this format the
colonies are used as a working reference library, and the master library is
spared frequent freeze/thaw cycles that may decrease culture viability and
increase the risk of contamination. You can access the colony library by simply
using the replicator to "pick" the colonies and transfer them to your test media
or solid substrate.
Usually there is enough colony transferred that you do not have to
wait for the cultures to grow.
•PICK CLONES FROM SCREENING
PLATES WITH 384 PIN REPLICATOR
Whole libraries can also be screened
in mass and colonies or plaques
"picked" in mass from Omni Trays
with the 384 pin MULTI-BLOT™
Replicator registered to a COLONY
COPIER™. This procedure is far
more efficient and superior to individually
picking colonies or plaques
with tooth picks. We re commend the
87v- 384 Replicator because of its larger
pin size.
