Domoic Acid ImmunoSepDomoic Acid ImmunoSepTM Introduction Methods that use immunoaffinity chromatography (IAC) for sample preparation or detection are becoming increasingly popular as tools in the analytical applications. Besides, there is the possibility to combine this technique with others instrumental techniques. Several applications of immunoaffinity chromatography sample preparation are described in literature, applications such as: drug, foods, environmental analysis, micotoxins, natural toxins, etc. The immunoaffinity columns are prepared by binding specific antibodies to specially activated solid support and packing the support suspended in aqueous buffer solution into a cartridge. The analytes in the extracts or fluid binds to the antibody, impurities are removed with washing buffer solution, and then the analites are eluted with a adequate solvent for elution. Further the analysis of eluates can be done by different techniques (HPLC, GC, Electrophoresis, etc.) ImmunoSepTM pack for Analysis of ASP toxins Amnesic shellfish poisoning (ASP) is caused by consumption of different species of shellfish that have accumulated domoic acid, a neurotoxin produced by diatom. The neurotoxin properties of domoic acid result in neuronal degeneration and necrosis in specific regions of the hippocampus. Monitoring programs are in place in numerous countries worldwide and closures of shellfish harvesting areas occur when domoic acid levels exceed regulatory limits. Control mechanisms have been introduced in order to carry out monitoring phytoplankton and shellfish products, and different analytical tools have been proposed for the control of this toxin. The high affinity and the high selectivity of the antigen antibody interactions for domoic acid and its isomers allow the specific extraction and the concentration of this toxic analyte previous to instrumental analysis by HPLC-UV or HPLC-MS.
