EvaGreen For qPCR
|
 |
We know that you have been looking hard
for an alternative.
Now... Look no further. EvaGreen is not
just another SYBR Green I, it is simply the best green fluorescent DNA
dye for qPCR |
|
Figure1. PCR kinetic plots using EvaGreen
(white square- and SYBR Green I (black triangle), respectively, in
titration of human genomic DNA. DNA amplifications with SYBR Green I
were carried out using a commercial SYBR Green I product with optimized
enzyme and SYBR Green I concentration.

Very Little PCR inhibition
Unlike SYBR
Green, EvaGreen shows very little PCR
inhibition.
Highly bright fluorescence
The fluorescence
brightness of EvaGreen in qPCR is
simply unmatched by that of SYBR Green I.
Superior stability
Unlike SYBR
Green, which has been reported to be unstable,1 EvaGreenΤΜ is extremely
stable.
Excellent compatibility
Simply replace
SYBR Green with EvaGreen. No need
to change anything else in your existing formulation and instrument
setting. The only difference you will see is a better performance. |
Biotium in collaboration with AlleLogic
Biosciences Corp. has developed EvaGreen as a superior fluorescent DNA
stain for quantitative real-time PCR (qPCR). EvaGreen is truely a
remarkable dye in many aspects. Upon binding to DNA, the fluorescence of
EvaGreen is several-fold higher than that of SYBR Green I while the dye
shows very little inhibition to the PCR process. Unlike SYBR Green,
which has been reported to be unstable, EvaGreen is highly robust, both
thermally and hydrolytically under alkaline or acidic condition. In
addition, the absorption and emission spectra of EvaGreenTM are similar
to those of SYBR Green I or FAM, which means that the same optical
setting for SYBR Green I can also be used for EvaGreen.
We now offer two EvaGreen products: 1)
EvaGreen at 20x concentration (#31000); and 2) EvaGreen qPCR Basic Mix
at 2x concentration (#31001). The Basic Mix contains everything you need
to run a qPCR except for the Taq enzyme. We will soon offer EvaGreenTM

EvaGreen and SYBR Green I at two different concentrations. SYBR Green I
exhibits significant PCR inhibition at 1x concentration (0D~0.05) while
EvaGreen does not (See inset) |

Figure 5. DNA melting curve analysis using
EvaGreen and SYBR Green I, respectively, with 4 different amplicons: 1)
TBP (--); 2) SDHA (--); 3) RPL4 (--); and 4) HMBS (--). SYBR Green I
exhibits significant PCR inhibition, resulting in occasional formation
of an extra melting peak. |
